范文编号:SW180 范文字数:10603,页数:26 附任务书,开题报告,文献综述,外文翻译 摘要:用萎缩芽孢杆菌(Bacillus atrophaeus)的芽孢来作为生物指标,以此来检测灭菌仪器的灭菌效果。国内外大多采用固体和液体培养法,本实验采用半固体培养基滤膜法来培养萎缩芽孢杆菌及其芽孢。这种方法既保持固体和液体培养法的优势又克服了它们的不足,同时提高萎缩芽孢杆菌芽孢形成率及细菌总数和芽孢数量,研究了营养条件对其芽孢产率的影响。结果表明:芽孢形成的最适培养基组成为(g/L):葡萄糖10g、酵母浸膏10g、NaC1 5g、MnSO4·H2O 0.6g;最适pH值为7.0。在上述条件培养下30℃恒温培养24h后,芽孢数量最高可达到 6.03×108mcfu/ml,芽孢形成率可达到82.59%。 关键词:萎缩芽孢杆菌 半固体培养 滤膜培养 芽孢形成率 Abstract: In order to detect the effects of sterilization equipment sterilization,using the spores of bacillus shrinking as biological indicators. Of nutritional condition on the spore yield. The results showed that:the optimal sporulation medium composition for(g/L):10g glucose、10g yeast extract、5g NaCl、0.6g MnSO4·7H2O、pH 7.0.Cultured under these conditions 30℃,after 24h incubation,the number of spores can reach 6.03×108mcfu/ml,sporulation rate reached82.59%.
