抗球虫药妥曲珠利中间体的合成

摘要：妥曲珠利(toltrazuril),简称为甲苯三嗪酮,属于三嗪酮类衍生物，其杀虫效果好,毒性低。妥曲珠利可用于鸡、兔、鸽、仔猪等畜禽的球虫感染,一般为溶液剂,饮水即可,使用方便,疗效确切。本文主要介绍其中间体3-甲基-4-(4-三氟甲硫基苯氧基)-苯胺的合成。通过文献综述比较而得到妥曲珠利中间体的合成路线，其中包括以4-三氟甲硫基苯胺为原料，采用重氮－水解羟基化法合成4-三氟甲硫基苯酚；4-三氟甲硫基苯酚和原料2-甲基-4-硝基氯苯缩合成3-甲基-4-(4-三氟甲硫基苯氧基)-硝基苯；在酸性溶液中用铁进行化学还原合成3-甲基-4-(4-三氟甲硫基苯氧基)-苯胺这三步反应，通过实验得到三步反应的收率分别为79.1%～80.8%、86.3%～88.1%、89.2%～91.0%，收率较高，已达到预期目标。

Abstract: Toltrazuril is Toluene triazone for short, belonging to triazone ramification, which has good insecticidal effection, low toxicity. Toltrazuril is used for chickens, rabbits, pigeons, pigs and other livestock and poultry which infected with coccidian. Commonly it is solution, just feeding by drinking water so it is convenience to use. This article mainly discuss the synthesis of its intermediates 3-methyl-4-[(trifluoromethyl)thiophenoxy]-aniline.Comparing the literature review we conceived the synthesis routes of toltrazuril intermediate by referring lots reference. This article used 4-[(trifluoromethyl)thio]-aniline as raw materials, adopting diazo-hydroxylation hydrolysis method to synthesze the 4-[(trifluoromethyl)thio]-phenol; 4-[(trifluoromethyl)thio]ph-
enol 2-methyl-4-nitro chlorobenzene condense 3-methyl-4-[4-(trifluoromethyl)thio-phenoxy]nit-
rophenyl;In acidic solution use chemical reduction of iron synthesis of 3-methyl-4-[4-(trifluoro-
meth-yl)thiophenoxy]-aniline. From the experiments we have got high productivity of three-step reaction, 79.1%～80.8%,86.3%～88.1%,89.2%～91.0% separately, which have got expected results.
