范文编号:SW021 范文字数:9104,页数:25 摘 要:本范文建立小儿咽扁颗粒中绿原酸含量的测定方法。采用HPLC法测定小儿咽扁颗粒中绿原酸的含量,采用依利特Hypersil ODS柱,乙腈-0.2%磷酸溶液(10∶90)为流动相,流速为1.0mL/min,超声和回流提取绿原酸的效果相当,但超声方法简单,故选择超声作为提取绿原酸的方法,以50%甲醇作为提取溶媒,提取时间为20分钟, 检测波长为327 nm。结果表明:绿原酸在1.016~101.6μg/mL之间呈良好的线性关系(γ=1),绿原酸含量测定方法,进样精密度良好,绿原酸在50%甲醇中12小时之内稳定,绿原酸含量测定方法,重复性及日间精密度良好,平均加样回收率为98.6%(n=6),RSD为0.55%。本法操作简便、易行、准确、重现性好,可用于该制剂的质量控制。 The Determination Of Chlorogenic Acid Content In Xiao’er Yan Abstract: This article is about establishing a method of quality control for assaying chlorogenic acid in Xiao’ er yan bian particle.Xiao’ er yan bian particle was analysized by HPLC.An yi li te Hypersil ODS column was used with a mobile phase consisted of mixture of acetonitrile—0.2%phosphoric acid (10:90,by volume ratio)by flowing rate of 1.0mL/min in this method.The methods of extracting chlorogenic acid by hypersound and backstreaming were equivalented.The hypersound was more convenient ,so we chose hypersound as the method to extract chlorogenic acid. Using 50% (by volume ratio) methanol as the menstruum,the time of extracting was 20 minutes.And VWD detector wavelength was 327nm.The results suggested that the linear range of chlorogenic acid was 1.016~101.6μg/mL (r= 1).The precision of sample introduction was very good by this method and chlorogenic acid was steady in 50% (by volume ratio) methanol in 12 hours . The determination reproducibility and daytime degree of precision of the method were very good.The average recovery was 98.6% (RSD=0.55%,n=6) .
