范文编号:SW112 范文字数:14869,页数:27 摘 要:本文是以海藻酸钠为载体,固定化醋酸菌发酵乙醇生产醋酸。研究了固定化细胞的制备,制备的颗粒的半径和体积大小。另外,还对接种环数、种子培养时间进行了优化。最主要的是研究了不同酒精初始浓度对乙酸产量、菌体个数、菌体生长周期、葡萄糖消耗量、乙醇消耗量的影响。最后我们得到如下结论:(1)固定化细胞颗粒平均直径为5.0mm,颗粒体积为65.42mm3;(2)优化接种数在15环、培养时间为36-48小时之间,发酵液菌体个数最多;(3)葡萄糖浓度与葡萄糖OD值的换算公式:y=4.7237x,单位为mg/ml;(4)菌体个数与菌液OD值的相关性:y = 402.88x,单位为107个/毫升;(5)酒精初始浓度对乙酸产量的影响:在各酒精浓度中,5%酒精浓度的发酵液乙酸转化率最高,最佳培养时间为96小时;(6)酒精初始浓度对菌体个数的影响:不同的酒精初始浓度影响着菌体的生长,酒精浓度越高对菌体的生长起着延长生长延迟期的作用;(7)酒精初始浓度对葡萄糖消耗的影响:乙醇的含量越高则葡萄糖的消耗就越少;(8)酒精初始浓度对乙醇消耗的影响:在72小时时间点,乙醇消耗完毕后,醋酸在72-96小时时间段的继续生成,是由于乙醛没随乙醇的消耗完毕马上转化为醋酸而引起的滞后反应。 关键词:固定化;乙醇;醋酸菌;醋酸钙镁盐 Abstract: This paper is based on alginate as a carrier of fixed acetic acid fermentation ethanol production of acetic acid.
