范文编号:SW161 范文字数:9965,页数:22 摘要:本文讲述从自然界的土壤中筛选耐高浓度丁醇的耐丁醇微生物。工业生产中,当丁醇的浓度达到11g/l时,50%的微生物都会因为丁醇的抑制而停止生长。通过梯度平板法筛选得到耐高浓度丁醇的微生物,并进行纯化分离。由于耐丁醇菌在固体培养基上的生长周期较长,所以需要进行一系列的液体培养,以确定所获得的耐丁醇菌是因为本身的生长周期原因抑或是固体培养基中的丁醇挥发。最终,确定筛选得到的耐丁醇菌的耐丁醇的浓度为60g/l。同时,绘制耐丁醇菌不加丁醇和加丁醇两种情况下的生长曲线,并进行耐氧性检测和一些简单的菌种鉴定工作,大致确定所获得的耐丁醇菌为芽孢杆菌属。最后,再进行斜面保藏,将菌种保存在冰箱中。 关键词:菌种筛选;丁醇;生长曲线;菌种鉴定 Abstract: This article says the way of screening the high-concentration butanol resistant strain from the soil in nature.In the industrial production, when the concentration of n-butanol reached 11g/l, because of the inhibition of n-butanol, 50% of the strain will stop the growth. Screening of the high-concentration butanol resistant strain through gradient plate method, and then separated and purificated the butanol resistant strain. Butanol resistant strain as a result of solid medium in the growth cycle is longer, so it is necessary to carry out a series of culture in the liquid medium to determine the reason is the growth cycle is longer or the volatile of n-butanol in the solid medium. Finally, screening of the butanol resistant strain to be resistant to the concetration of 60g/l. Meanwhile, the growth curves of the strain in the liquid medium with and without n-butanol were draw, and do the oxygen resistant detection and some simple strain identification, more or less determined the butanol resistant strain is Bacillus sp.
