范文编号:SP118 范文字数:11376,页数:23 摘 要:硫代葡萄糖苷是植物的一种含硫次级代谢产物,具有抗癌活性,其降解物是异硫代氰酸盐(isothiocyanate,ITC)。苯甲基异硫氰酸盐(benzylisothiocyanate,BITC)和苯乙基异硫氰酸盐(phenzylisothiocyanate,PEITC)是生物活性化合物,能够有效的预防肺癌等。但常温下其化学结构不稳定,易挥发,受光、热、氧等条件影响。本实验以β-环糊精为壁材,对β-环糊精-BITC和β-环糊精-PEITC的包埋工艺进行了探讨研究,希望能增加BITC和PEITC的稳定性。实验以包埋率和包埋度为主要包埋参数,选取芯材与壁材质量比、包合温度、包合时的搅拌速度为影响因素,按L9(33)正交表进行了正交试验,优化包埋工艺;另外,本实验建立了用紫外分光光度法定量测定异硫氰酸盐的方法。实验结果表明:三因素对BITC和PEITC包埋效果的影响不同,以包埋率为指标,β-环糊精- BITC和β-环糊精-PEITC最佳包埋条件分别为:A3B3C1 和A1B3C3;以包埋度为指标,BITC和PEITC的最佳包埋条件分别为:A1B3C3 和A1B2C2。 Abstract: Isothiocyanate(ITC) is the degradation product of glucosinolate (GS) which is secondary metabolite including sulfur with anti cancer activity. Benzylisothiocyanate(BITC) and phenzylisothiocyanate(PEITC) are two important biological activity compounds with remarkable anticancer activity. But they are unstable in normal temperature and easy to volatilize and destroyed by light, heat and oxygen, and so on. β-Cyclodextrin (β-CD) was selected as wall-material and the embedding technology of β-CD –BITC and β-CD-PEITC was studied. TO investigate the effects of different quality ratio of ITC and β-CD, inclusion temperature and the agitation speed on inclusion ration and inclusion degree, an orthogonal test comprising nine test runs based on these factors at three different concentrations were carried out. The quantificative analysis method was established. The results showed that the optimum conditions were A3B3C1 and A1B3C3 with embedding ratio as objection, while A1B3C3 and A1B2C2 with embedding degree as parameter.
