范文编号:SW013 范文字数:8707,页数:23 摘 要:本范文主要介绍了双孢蘑菇的研究进展(包括双孢蘑菇的营养价值与经济意义),多酚氧化酶活性及性质,并测量了双孢蘑菇的褐变度及其对应的多酚氧化酶(PPO)活性的变化。褐变度是指双孢蘑菇采摘后在一定温度下储藏不同时间后的褐化程度,它是用测色色差计来测量的;本试验主要以L值大小作为双孢蘑菇褐变程度的指标,L值越大,表示颜色越白,褐变越轻。PPO催化各种酚与O2氧化为醌。本实验是采用邻苯二酚为底物,在磷酸钠缓冲液PH6.5的反应体系中,PPO催化邻苯二酚形成褐色的醌,在分光光度计410nm处使反应体系的OD值产生变化,通过OD值上升的读数变化确定PPO的酶活大小。结果表明,双孢蘑菇内的PPO参与了褐变过程,并与褐化程度有一定的关系。 关键词:双孢蘑菇;褐变;多酚氧化酶 Study on the relationship between agaricus bisporus browning and activity of polyphenol oxidase Abstract: This article introduces the progress of agaricus bisporus(including the nutritional value and economic significance of agaricus bisporus), polyphenol oxidase activity and nature, and measured the agaricus bisporus browning and its corresponding level of polyphenol oxidase (PPO) activity .Browning is agaricus bisporus which after picking under a certain temperature storage at different times after the extent of browning,it is used color measurement to measure , the size of L is degreed the agaricus bisporus browning in this test, L greater value, as the color greater white and as the browning lighter. PPO catalytic various of phenol and O2 oxidation of quinine. This study was used as the substrate catechol, sodium phosphate buffer pH6.5 response system, PPO catalytic formation of catechol brown quinone, Spectrophotometer at 410nm in reaction to the OD values change, use the reading of OD value rose to determine the size of PPO activity.
