范文编号:SW029 范文字数:6631,页数:22 摘 要:本文从雷帕霉素的摇瓶发酵着手,对斜面培养基、种子培养基和发酵培养基的组成和浓度进行优化,同时考察了种子菌龄、摇瓶收获时间和HPLC萃取条件。实验主要采用单因素法来比较各因素的影响,以达到优化的目的。雷帕霉素的测定方法有生物活性测定法、薄层层析法和液相色谱法,经过比较得出,高效液相色谱法可以简洁的定量分析雷帕霉素,而且测定结果比较可靠。在对培养基和各种条件进行优化后,雷帕霉素的产量得到了很大的提高,在优化后的条件下雷帕霉素的产量达到140mg/L,为进一步工业化生产雷帕霉素提供了基础。 关键词:雷帕霉素;免疫抑制剂;高效液相色谱;发酵条件优化 Optimization of fermentation conditions for production of Rapamycin Abstract: In this paper, the rapamycin fermentation to proceed on the slant medium, seed medium and fermentation medium to optimize the composition and concentration, and seed strains studied age, harvest time and shake-flask extraction HPLC conditions.
