范文编号:SW155 范文字数:11122,页数:29 摘 要:本范文通过对出发菌株运动酵母单胞菌进行耐酸性试验使其能在酸度较高的环境中存活,然后对耐酸性试验得到的耐高酸菌株进行物理化学诱变,如紫外-EMS(甲基磺酸乙酯)联合诱变,加热诱变,紫外-NTG(亚硝基胍)联合诱变,并利用TTC加层平板法和CM,MM平板法筛选到一株高产的丙酮酸菌株。然后对筛选得到的高产菌株进行摇瓶发酵,并利用HPLC法,2,4-二硝基苯肼法,硝酸铁法测得发酵液中丙酮酸的含量。 关键词:丙酮酸 诱变 HPLC 2,4-二硝基苯肼 硝酸铁 Abstract: In this article, movement of the Zymomonas mobilis Aeromonas acid test to enable them to high acidic environment to survive, and used the strains after the acid test of the high acid-resistant strains to physical and chemical mutagens such as UV-EMS ( ethyl methane sulphonate) mutagenesis combined heat mutagenesis, UV-NTG (nitrosoguanidine) mutagenesis joint and increase the use of TTC plate layer and the CM, MM filter plate to a high yield of acetone acid strains.
